چکیده

For investigation of the properties of smooth muscle contraction hypertonically added 40 mm K medium has been used as a routine in this laboratory. Others have used isotonic high K/Na deficient medium for the same purpose. The distinct difference xisting between the mechanical response of guinea pig taenia coli in hyper-40 K medium and that in iso 152 K medium has been reported earlier. The response in hyper-40 K medium consist ed of a phasic and a tonic phase (1). The tonic response in hyper-40 K medium at a steady level was very much higher than that in iso-152 K medium (2). The tissue calcium in hyper-40 K medium increased con siderably above control levels while that in iso-152 K medium decreased sharpely below control levels (2). The "Ca uptake of the fraction which did not exchange within 4 minutes in hyper-40 K medium was also considerably above control while that in iso-152 K medium was not above control (2). The tonic response in hyper-40 K medium has been found to be abolished by factors suppressing sodium transport mechanism or aerobic breakdown of carbohydrates (3), but the nature of this response in iso-152 K medium has not been studied. This small contracture was therefore investigated . Strips of taenia coli isolated from white male guinea pigs were suspended in an organ bath containing Tyrode solution of the following composition (mm) : NaCI 136.8, KCl 2.7, CaCl, 2.5, MgC1, 1.0, NaH,P04 0.4, NaHCO3 11.9 and glucose 5.5, saturated with 95% 02 and 5% CO, mixture at 37°C. Iso-152 K medium was prepared by substituting all sodium ions of normal Tyrode solution by potassium ions. Hyper-40 K me dium was prepared by the addition of KCI crystals to normal Tyrode solution to give a final concentration of 42.7 ms K. Tension changes were recorded isometrically with mechanoelectro-transducer.

Increase in membrane permeability in the absence of Ca and Mg in the smooth muscle of guinea-pig taenia coli.

In Ca-deficient smooth muscle of guinea-pig taenia coli, repeated application of high-K solution (45.4 mM) containing Ca induced contractions of similar shape and magnitude. In muscle treated by a Ca- and Mg-deficient solution, however, addition of Ca and K either did not induce contraction or induced only a delayed contraction. Ouabain (1 X 10(-3) M) also inhibited Ca- and K-induced contractio...

Changes in tension Ca movement and metabolism of guinea pig taenia coli in varying concentrations of external Na and K.

In a previous report (1) it was shown that in guinea pig taenia coli hypertonically added 40 mm K induced a large tension and that both tissue Ca and "Ca uptake of the muscle measured after a 4 minutes wash increased, while in 152 mm K/0 mm Na medium only a small tension remained and '5Ca uptake did not increase while tissue Ca decreased . It has also been shown that the tonic response in hyper...

Sodium in smooth muscle relaxation.

The contraction of guinea-pig taenia coli due to high K+ could not be reversed by washing when Na+ was absent from the medium. Reintroduction of Na+ (7 mM or more) caused relaxation. Similar results were obtained with rat uterus. The effect of sodium replacement was not due to change in ionic strength because equal or higher osmoles of choline, Ca++, K+, Mn++, or Mg++ had little or no effect. P...

On the inhibitory mechanism of bassianolide, a cyclodepsipeptide, in acetylcholine-induced contraction in guinea-pig taenia coli.

The effect of bassianolide (BASS), a cyclodepsipeptide, was investigated on mechanical response, membrane potential, intracellular Na and K contents and 45Ca uptake in response to acetylcholine (ACh) in guinea-pig taenia coli. BASS (10(-5) M) as well as verapamil (5 X 10(-7) M) and papaverine (3 X 10(-5) M) selectively inhibited the tonic component of the contraction induced by ACh (10(-5) M), ...
